Trifluridine (trifluorothymidine, TFT), a fluorinated pyrimidine nucleoside, is an anti-herpesvirus agent and an antitumor antimetabolite agent. It is an analog of thymidine which inhibits thymidylate synthase possesses antiviral and anticancer activity. After phosphorylation by thymidine kinase, it is incorporated into DNA where it induces DNA-damage and interferes with repair enzymes. Enhances frame shift insertion and deletion in CRISPR genome editing in pluripotent stem cells.

1. Trifluridine is used as anti-herpesvirus antiviral agent in ophthalmie preparations.
2. Trifluridine is also used as an anti-cancer nucleoside, mainly for the treatment of colorectal cancer, especially for the early treatment of colon cancer patients. Trimethoprim interacts directly with the DNA of the cancer, preventing the DNA from functioning properly.
3. Trifluridine ia a broad-spectrum antiviral drug, inhibits herpes simplex virus, adenovirus, herpes zoster virus, HSV-I, HSV-II, CMV, varicella virus and some adenoviruses.
4. Trafluridine is mainly used for herpes virus causing conjunctivitis, keratitis and other herpetic eye diseases. It is more effective than iodoside. It is commonly used in patients who have failed to respond to cytarabine treatment.
